Received free from my pool table seller without instructions. As a newcomer in billiards, can anyone tell me how to use this by applying to my hand?

Welcome to the world of Billiards. It is simply a rub on chalk, after the protective plastic is removed just rub your hand over the cone like you were picking up your favorite beverage. Some players apply to both hands and some apply to just the forward hand that the cue slides through. Enjoy!

how do you clean it out of carpet?

I don't get it on the carpet but if I did I'd vacuum. It's just white chalk.

Will it fit on a paper towel dispenser?

Hello. You may be able to fit this on to some paper towel dispensers, assuming that the dispenser is a vertical style and post is small enough.
